About Composition
Diclofenac:Diclofenac is phenylacetic acid derivative and non -steroidal anti -inflammatory drug Diclofenac inhibit cox -1 and cox-2 enzyme and prevents the synthesis of prostaglandins.
Paracetamol:
Paracetamol is known as acitominophen which is a pain releiver and fever reducer.
Mechanism of Action
Diclofenac inhibits the synthesis of prostaglandins by inhibiting the cox-1 and cox-2 enzyme and prevents the formation of PGG2 which the precursor for the synthesis of other prostaglndines.
Paracetamol is a weak inhibition of COX-1 and COX-2, further inhibiting prostaglandin synthesis.
Pharmacokinetics
Absorption: Both the drugs are orally absorbed from the GIT.
Protein Binding: Diclofenac is mainly bound to albumin whereas paracetamol do not show protein binding.
Metabolism: Diclofenac undergoes oxidation and conjugation.
Excretion: Both are excreted via urine.

Contraindication
- Contraindicated in third trimester of pregnancy
- Contraindicated in hypersensitivity.
